Tip #203: Empower User Question: Comparing UV Spectra from Different Channels in a Report, at the Same Retention Time
Welcome back to Get Empowered! In the last Empower tip-of-the-week post for Empower Chromatography Data System, we revisited the VLOG series showcasing Empower as an important tool to implement Method Lifecycle Management (MLCM) and discussed using the eCord with ACQUITY and ACQUITY Arc systems which automates column knowledge management and serves as a control strategy for column usage (Episode #9 / Tip #202).
In this week’s tip, we will answer another question from an Empower user regarding the overlay of UV spectra.
Q: How do we compare UV spectra from different channels in a report?
Another great question.
Let’s get started.
Please Note: For this tip, we will overlay spectra from both channels of data at the same retention time.
Highlight the 3D channels, Right click and select ‘Preview/Publisher’ (figure 1).
When the ‘Open Report Method’ dialogue opens, select ‘Use the currently opened Report Method named Untitled’ and click ‘OK’ (figure 2).
Right click in the empty space on the right side of the window and select ‘Method Properties’ (figure 3).
Under ‘Report Type’, select ‘Summary By All’ and click ‘OK’ (figure 4).
On the left side of the window, expand ‘PDA/MS’ groups, expand ‘Extracted Spectral Plots’ and double click on the group called ‘254nm Extraction Group’ (figure 5).
Right click on the group and select ‘Extraction Plot Properties’ (figure 6).
On the ‘Chromatogram’ tab, delete ‘line 1’ (figure 7).
On the ‘Spectra’ tab make the changes as shown in Figure 8 and enter the appropriate ‘extraction time’ for the spectrum (figure 8).
On the ‘Overlay’ tab, select either ‘Make Stack Plot’ or ‘Overlay in Single Plot’ (figure 9).
On the ‘Legend’ tab, delete the ‘Plot Title’ (figure 10).
Expand ‘Sample’ (figure 11).
Double click on ‘SampleName’ (figure 12).
You now have an overlay of spectra from two different 3D PDA channels at the same retention time (figure 13).
