Tip #201: Empower User Question: Calculation of USP Signal-to-Noise Ratio

Welcome back to Get Empowered! In the last Empower tip-of-the-week post for Empower Chromatography Data System, we answered an Empower user question about identifying peaks and celebrated a milestone with Empower Tip #200.

In this week’s tip, we will answer another question from an Empower user regarding the calculation of USP signal-to-noise ratio.

Q: Why am I not getting the USP signal-to-noise calculated when I use a ‘blank’ for the noise calculation? 

Another great question.

Let’s get started.

Please Note: For this tip, I am using generic data as the customers data is confidential.

Step 1

The result displayed does not have the USP signal-to-noise (s/n) nor the USP Noise calculated (figure 1).

Step 2

The ‘Suitability’ tab in the Processing Method shows the correct selection for calculating USP signal-to-noise (s/n) and the use of a blank injection to calculate noise (figure 2).

Step 3

Working backwards, ‘Alter Sample Set’ displays the ‘Sample Matrix’ field.  The selection for the ‘Blank’ is different from the rest of the samples and that is the problem (figure 3).

Step 4

Modify the ‘Sample Matrix’ to be the same for the blank (figure 4).

Step 5

Reprocess the Sample Set and now the result for the same sample shows the values are calculated (figure 5).

Step 6

Search on ‘Verifying System Performance’ in the online help and you will see a Tip regarding the Sample Matrix (figure 6).
